If you edit them in the file you have to edit them straight after in the ACP. If you edit them via phpMyAdmin you'll still have to delete settings.php to get it to pull from the database again. If you edit settings.php and then delete it, it'll just regenerate from the database again so they'll still be wrong. MyBB will only re-write settings.php when you save the settings, it won't touch it otherwise. If you edit settings.php and still can't login, chances are you've not set them right, but either way, editing the file is less hassle than doing it via phpMyAdmin.
