is there any way to not count user post in selected forum,
and any way to do this- Need minimum 40 character to be counted as post.
please help.

For your first question, it's a core MyBB feature.
Edit the forum settings for the forum in question. You should see a setting for "Yes, posts in this forum should count towards user post counts". Make sure the box is NOT checked.

If you mean in all forums then it's again a core feature (Settings->Posting->Minimum Message Length).
