Please tell me how to delete/remove all or multiple smiles at once.

ACP > Configuration > Smilies > Mass Edit

Tick all the checkboxes at right which you wanna delete and hit "Save Smilies".

There are up to 5000 smiles on my forum. Is there any method to delete them at once ?

Try running this query:

DELETE FROM `mybb_smilies`
