Could someone assist or explain how I would change the format of the URL to go to someone's profile.

Example:

From...
http://www.dummyaddress.co.uk/en/member....file&uid=1

To...
http://www.dummyaddress.co.uk/en/member....on=profile&username=Knerba

You can simply use this (works only in postbit template): {$post['profilelink']}
Install SEO plugin if you don't have it yet!
