I want to change the label "Username". What file is that label located in?

Do you want to change it on the register form or login form?

You should be able to find and change it by going to

Admin CP -> Languages -> English (America) -> global.lang.php

Just search the page for “username” you should be able to find it and change it.

I want to change it on the registration form. Those entries in global.lang.php, are they for both registration and login?

Try and see that's the only way you'll find out but if it's global then probably yes.
