It should although I don't believe it's fully tested. From my understanding it shouldn't be a problem though.

Yes, the MyBB comm. forums here were running on lighttpd during the first few weeks of the MyBB 1.4 release to cope with the load.
The only thing that won't work is your .htaccess SEO friendly urls and lighttpd doesn't support mod_rewrite. It can however be rewritten if you look up a few tutorials on how to do it.
