Is there any way that if a user only replies with a smilie or multiple smilies that it won't count toward their overall post count?

No, because the smilies are still characters. E.g, you have to write something for the smiley to actually show up anyway. So if you tried blocking out that text; you'd most likely block it all out == no posts counting anywhere.

Couldn't you just set posts so that they have to be a over a certain number of characters?

Now people could just post multiple smiles, but it will force those who want to put post one smile to also post some text.

Yeah, using something like LaBrocca's plugin the Minimum Post Suite or something you could have a minimum amount of text. So they wouldn't just be able to post a smilies or whatever.

Admin CP -> Configuration -> Settings -> Posting:

- The first thing you see:
Minimum Message Length
The minimum number of characters to post.
- I have mine set to three, I suggest changing yours to 4-6.
- Scroll down and press save settings.
