First question - How can I make it so the user has to use a prefix for a thread and if they attempt to post a thread without selecting a prefix, it will stop them and give them a simple error.

Second Question - I installed the Profille Comments mod for 1.6, and I was wondering how I can expand the comments box and the comments that appear below it. Basically just increase the width of the comments box and comments that appear below the box.

For your first question, that would require a plugin.
For your second question, you will need to go to the Profile Comments thread and request support there.
