What file am I looking for to edit if I want to make a text change to the page with the user registration form? I want to add something to the box labeled [username:] with additional requirements.

You don't have to edit any file. Just go to the ACP -> Templates & Style -> Templates -> select your template set (normally Default Templates) -> Member Templates -> member_register.
